It is better to get vaccinated than to "hem"

In Chile, they invented an inoculation against alcoholism

Copper newsChilean scientists have applied to the treatment of chronic alcoholism one of the varieties of gene therapy – the method of antisense therapy.

A single injection of the "anti-alcohol vaccine" developed by them for six months to a year causes symptoms of a severe hangover immediately after taking even a minimal portion of alcohol due to blocking the process of its metabolism in the liver, reports The Daily Mail (Alcoholism vaccine will give drinkers an immediate hangover if they drink even a small amount of booze).

Antisense therapy is based on stopping the synthesis of a key protein responsible for the development of the disease by suppressing the expression of the corresponding gene.

This is achieved by introducing specially designed short nucleotide sequences (antisense RNAs, ac-RNAs), complementary to the target protein matrix RNA.

Developed by a group of specialists under the leadership of the head of the Institute of Cell Dynamics and Biotechnology of the University of Chile, Dr. Juan Asenjo, ac-RNA blocks the synthesis of the enzyme acetaldehyde dehydrogenase in the liver, so that the process of decomposition of ethyl alcohol to carbon dioxide and water stops at the stage of acetaldehyde (acetic aldehyde), which is a tissue poison and causes acute intoxication of the body, accompanied by symptoms such as nausea, vomiting, tachycardia, general malaise, and so on.

The effect of inhibiting the expression of the acetaldehyde dehydrogenase gene, as reported by the authors, persists for six months to a year and is irreversible throughout this period.

According to Asenyo, in their work, his group focused on a well–known phenomenon - about 15 to 20 percent of the population of the countries of the Far East, such as Japan, China and Korea, carries a genetic mutation similar to an artificially induced "anti-alcohol vaccine". These people also have blocked the synthesis of acetaldehyde dehydrogenase in the liver, which is why they cannot drink alcohol.

The authors plan to begin preclinical testing of their development in mice in February to develop the correct dosage, and in November its first clinical trials should begin in India. Aseno predicts that, if successful, the "anti-alcohol vaccine" could be released to the market in two years.

According to the World Health Organization data for 2011, one in fifteen Chileans suffers from alcohol addiction.

It is worth noting that the idea of using pharmacology methods to combat alcoholism is not new. For example, the drug disulfiram, developed almost a hundred years ago, acts similarly to the "anti-alcohol vaccine", inhibiting the production of acetaldehyde dehydrogenase in the liver and causing intoxication with acetic aldehyde. The duration of action of one tablet is 48 hours, implantation is used to create a constant concentration of the drug in the body. However, the drug has many side effects and contraindications, which limits its use.
