Synthetic opioids are better than morphine

A new "gold standard" of anesthesia is being born

Sergey Syrov, XXII CENTURY

Drugs that can become the new "gold standard" of anesthesia have been synthesized. You can read about the development of American pharmacists in an article available online (Zadina et al., Endomorphin analog analgesics with reduced abuse liability, respiratory depression, motor impairment, tolerance, and glial activation relative to morphine), a paper publication in the journal Neuropharmacology is scheduled for June 2016. We are talking about four peptides capable of overcoming the blood-brain barrier and interacting with opioid m-receptors.

The resulting substances are similar in structure to endomorphins and have a long-term analgesic effect similar to known opioids, but with significantly weaker side effects. In a series of animal experiments, new drugs were compared with morphine. In doses providing an analgesic effect equal to morphine, the studied peptides showed an unprecedented low level of negative side effects: there was no respiratory depression, no impaired coordination of movements, or the formation of dependence.

Researchers believe that the low level of harmful effects of new drugs is due to the fact that they work with neuroglia more selectively than morphine, which acts, in particular, on the glial receptors p38, CGRP and P2X7, which determines its narcotic properties.

The work on the synthesis and testing of drugs was carried out at the Faculty of Medicine of Tulane University in New Orleans, USA (Study: New drug could be safer, non-addictive alternative to morphine – VM). Clinical trials of new painkillers will begin in the next two years.
